He was the son of Thomas Johnston, Jr., who was born in about 1750, in Ireland and Sarah Stapleton, who was born in 1749, who was born in Ireland.

Samuel Johnson, Sr. was married to Sarah Pennington, in 1799, at Yadkin Rover, Rowan County, in the State of North Carolina. She was born on the Twenty-forth day of November, in 1776, at Wilkes County, in the State of North Carolina. They were the parents of:

James Johnston or Johnson was born on the Thirteenth day of July, in 1799, in the State of North Carolina and died on the Twenty-second day of December, in 1889, in the State of Kentucky. He was married to Margaret Gay.

Jane Johnson was born in about 1804, at Grayson County, in the State of Virginia and died on the twelfth day of December, in 1858, at Breathitt County, in the State of Kentucky.
